چکیده

We have reported elsewhere the racemization of lysine by cell preparations of Proteus vulgaris strain ATCC 4669 (Huang et al., 1958). The simplest explanation of the results presented is that Proteus vulgaris contains a lysine racemase. The distribution of this activity in bacteria has now been investigated. Almost a hundred species, from eleven families, have been examined. Only nine species, all from the genera Proteus and Escherichia, could be shown to catalyze the racemization of lysine. We have also examined the effect of Proteus vulgaris on alanine, glutamic acid and methionine. Only alanine was found to be racemized to an appreciable extent.

Racemization of lysine by a pseudomonad.

The recent paper by Huang and Davisson (J. Bacteriol., 76, 495, 1958) on the distribution of lysine racemase in bacteria reports its occurrence in only a few species of Escherichia and Proteus. Various other bacteria including 22 strains of various genera of the Pseudomonadaceae did not possess such activity. I wish to report evidence for the occurrence of lysine racemase activity in a Pseudomo...

Functional classification of amino acid decarboxylases from the alanine racemase structural family by phylogenetic studies.

Arginine decarboxylase (ADC) and ornithine decarboxylase (ODC) are involved in the biosynthesis of putrescine, which is the precursor of other polyamines in animals, plants, and bacteria. These pyridoxal-5'-phosphate-dependent decarboxylases belong to the alanine racemase (AR) structural family together with diaminopimelate decarboxylase (DapDC), which catalyzes the final step of lysine biosynt...

Analysis of the peptidoglycan of Rickettsia prowazekii.

In the present study, peptidoglycan from Rickettsia prowazekii, an obligate intracellular bacterium, was purified. The rickettsial peptidoglycan is like that of gram-negative bacteria; that is, it is sodium dodecyl sulfate insoluble, lysozyme sensitive, and composed of glutamic acid, alanine, and diaminopimelic acid in a molar ratio of 1.0:2.3:1.0. The small amount of lysine found in the peptid...

D-lysine catabolic pathway in Pseudomonas putida: interrelations with L-lysine catabolism.

The isolation of several mutant strains blocked in l-lysine degradation has permitted an assessment of the physiological significance of enzymatic reactions related to lysine metabolism in Pseudomonas putida. Additional studies with intact cells involved labeling of metabolic intermediates from radioactive l- or d-lysine, and patterns of enzyme induction in both wild-type and mutant strains. Th...

Crystal Structures of Lysine-Preferred Racemases, the Non-Antibiotic Selectable Markers for Transgenic Plants

Lysine racemase, a pyridoxal 5'-phosphate (PLP)-dependent amino acid racemase that catalyzes the interconversion of lysine enantiomers, is valuable to serve as a novel non-antibiotic selectable marker in the generation of transgenic plants. Here, we have determined the first crystal structure of a lysine racemase (Lyr) from Proteus mirabilis BCRC10725, which shows the highest activity toward ly...
